Determination of Genetic Variability using PCR-RAPD and Enzymatic Changes of Induced Callus in Two Rice Seed Embryo Lines under the Influence of Physical and Chemical Factors

Abstract

This study was conducted at the Agricultural Engineering College, Baghdad University, from 2021 to 2023. Tow callus lines, black rice and Amber-33 were used to induce genetic variability by UV-C radiation for 15, 30, 45 and 60 min. The callus grown on LD50 was cultured in different concentrations of polyethylene glycol (0, 1.5, 3, 4.5, and 6%). Two primers (0PB10 and 0PB 06) were used. The primers showed a difference in the number of bands after migration on agarose gel. The first primer showed the best result at 30 and 45 min duration for the black rice, which gave six bands, while for the Amber-33, the best result was at 30 min duration with nine bands. The second primer gave seven bands for the black rice at the 45 and 60 min doses, which were the best duration doses, while the same primer showed the best results at a dose of 15 min, giving six bands for Amber-33. The enzymes superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) were extracted. The 1.5% PEG concentration outperformed the other concentrations, giving the highest values for the enzymes SOD, POD and CAT of 7.09, 2.54, and 9.17, respectively.
